We found that OsP2 is not an MFS protein, and that this channel has a unique mechanism for regulation. We hypothesize that OsP2 is an osmosensing protein that plays a role in the accumulation of M. xanthus energy reserves. In the first aim, we will define the molecular characteristics of OsP2 by identifying the type of amino acids that make up OsP2, and by cloning and sequencing OsP2 cDNA. In aim 2, we will characterize the channel properties of OsP2 using electrophysiological techniques, and determine if there is a relationship between the OsP2 potassium channel and the function of energy reserves. In aim 3, we will characterize the role of OsP2 in the accumulation of energy reserves by expressing OsP2 in osP2 mutant cells and determining the accumulation of energy reserves in cells with impaired OsP2.
